目的 黄酮类化合物活性较高,但在甘草中提取率较低,难以满足市场需求。为进一步探究萃取甘草中黄酮类化合物的工艺条件,选取襄阳产甘草提取物中的黄酮萃取部分进行研究,采用单因素实验、正交实验与验证实验,总结黄酮的最佳提取工艺。方法 通过酶解-超声联用法提取甘草中的黄酮,采用分光光度法测定黄酮的含量,分别采用不同温度、pH、超声时间、纤维素酶处理时间进行实验,分析不同条件对甘草中黄酮提取率的影响,根据单因素实验数据,开展正交实验优化实验条件,然后开展验证实验。结果 襄阳产甘草中黄酮的最佳提取工艺条件为pH5、酶处理时间2 h、温度40℃、乙醇体积分数80%、超声时间30 min。结论 最佳提取方法稳定可靠,有利于甘草中黄酮类成分的提取。
Abstract:ObjectiveFlavonoids have high activity but have a low extraction rate in licorice, making it difficult to meet market demands. To further explore the extraction process conditions of flavonoids in licorice,the flavonoid extraction part from the licorice extract produced in Xiangyang was selected for research. Single-factor experiments, orthogonal experiments, and validation experiments were conducted to summarize the optimal extraction process of flavonoids. Methods Flavonoids were extracted from licorice using the enzymaticsonication combined method. The content of flavonoids was determined by spectrophotometry. Experiments were carried out with different temperatures, pH values, sonication times, and cellulase treatment times. The effects of different conditions on the extraction rate of flavonoids in licorice were analyzed. Based on the data from single-factor experiments, orthogonal experiments were conducted to optimize the experimental conditions, and then a validation experiment was carried out. Results The optimal extraction process conditions for flavonoids in licorice produced in Xiangyang were pH 5, enzyme treatment time 2 hours, temperature 40℃, ethanol volume fraction 80%, and sonication time 30 minutes. Conclusion The optimal extraction method is stable and reliable,which is conducive to the extraction of flavonoid components in licorice.
